CA-125, also known as Mucin 16 or MUC16, is a protein encoded by the MUC16 gene. MUC16 is a member of the mucin family glycoproteins. It is found to be elevated in the blood of patients with some types of cancer, most notably ovarian cancer.

You can eat and drink normally before the test, requiring no special preparation.
Your diagnostic lab may ask for your clinical history. Thus, you should carry your medical history and doctor’s prescription with you.
What to expect during the CA 125 test?
It is a fairly simple diagnostic blood test. A blood sample is taken from the vein in your arm or hand by the following steps-
- A tourniquet (elastic band) may be tied to your arm.
- The phlebotomist will locate your blood vessel.
- The site is cleaned with spirit.
- A Needle is inserted into your vein to draw the blood.
- A blood sample is then withdrawn into a syringe or a vial.
- Cotton is placed on the puncture site with mild pressure till the bleeding stops.
- A small band-aid is placed, which may be removed after an hour.
The entire procedure takes only a few minutes.
Are there any risks?
It is a very simple and safe test. The risks involved are the same as that in any other blood test. These include-
- Infection at the puncture site if proper sterilization protocol is not followed and the needle is contaminated. This is, however, very rare. You may ensure that your phlebotomist takes out the needle in front of you from a sterile pack.
- Multiple pricks if the blood vessel is not easily found
- Excessive bleeding at the puncture site- Please inform your healthcare provider if you are taking any blood thinners or have any bleeding disorders.
- Light-headedness due to stress or heavy bleeding.
Minor pain and bruising that might occur will resolve on their own in a very short time.

Does a normal CA 125 level necessarily mean I do not have ovarian cancer?
In about 50% of women with early-stage ovarian cancer, CA 125 levels may NOT be elevated. Similarly, 15% of women with advanced stage of the disease do NOT show an increase in CA 125 levels.

No.
Though high CA 125 levels are found in ovarian cancer, it can also point to other diseases.
Elevated CA 125 levels may be found in –
- Liver diseases
- Pancreatitis (Inflammation of the pancreas)
- Uterine fibroids (benign tumours of the uterus)
- Appendicitis (infection of the appendix)
- Endometriosis (a disorder where tissue similar to the inner lining of the uterus grows outside the uterus)
- Pelvic Inflammatory disease (an infection of female reproductive organs)
- During the first trimester of Pregnancy
- Other malignancies (cancer) like that of the uterus, fallopian tube, peritoneum, etc
An increased level may also be found in 1% of healthy individuals.
Hence, other investigations along with clinical signs and symptoms are correlated to come at a definite diagnosis.
